Mouse DNA preparation for Illumina chips


Preparation of samples for Illumina Mouse Medium Density Panel

 

SeqLab needs 5 ul of DNA at a concentration of 50 ng/ul (total 250 ng) for Illumina Mouse Medium Density Panel. Bring your samples on a ABgene 96-well PCR-plate, filling up the first two columns of the plate for each set of 16 samples. High quality DNA is important, and fragmented DNA does not yield good results.

 

  1. Extract DNA using for instance the Qiagen Blood and Tissue Kit (cat. no. 69504, 50 samples) . This kit results in DNA pure enough to use in the assay.
  1. Measure the concentration of your extracted DNA-samples (e.g. with Nanodrop).
  1. Lyophilize your samples if they are too dilute for 50 ng/ul dilutions:
    1. Calculate how much of your sample you need to take to get 500 ng DNA.
    2. Lyophilize the calculated amount.
    3. Resuspend the lyophilized DNA in 10 ul of molecular biology grade water overnight.
    4. Mix by pipetting or briefly vortexing the sample before continuing.
  1. Pipette 6 ul of your samples onto a ABgene 96-well PCR-plate, filling up the first two columns. Take your plate to SeqLab along with a plate map indicating sample names.
